Chitosan is an abundant natural polysaccharide that has several medical applications. It is a polymer of glucosamine that is partly acetylated and can be biodegraded in vivo. We have developed particular formulations of chitosan that can be used in conjunction with blood, cells and other biological components in order to repair tissues and have other therapeutic effects. Our research in chitosan is both fundamental and applied. In the fundamental area we are studying the polyelectrolyte behavior of chitosan in solution and in particular examining its interesting thermal gelation upon heating. The molecular mechanisms involved in gelation and other physicochemical phenomena are being investigated both experimentally and with theoretical mathematical models of polyelectrolytes in solution. The physical and biological properties of chitosan are very dependent on its molecular weight and distribution of acetyl groups. Therefore our research efforts also involve development of processes to control molecular weight and acetylation and the development of analytical methods to precisely characterize these two parameters.

Cartilage and Meniscus Repair

The knee joint and other joints in the body are organs composed of tissues such as cartilage, meniscus, ligaments, tendons and bones that must all function together. Unfortunately traumatic injury to these tissues and other disease processes can damage them to the point of limiting joint motion and causing pain, ultimately leading to arthritis and dysfunction. We are studying the structure, biomechanics and biology of articular cartilage and meniscus and using chitosan based biomaterials to develop means of regenerating these tissues when they are damaged. Our approach relies on formulating mixtures of chitosan solutions and blood components that can be applied to cartilage and meniscus lesions and stimulate their repair by activating host cell recruitment, vascularisation and synthesis of an effective repair tissue. We have succeeded in improving the repair of articular cartilage in animal models and this technology has been translated to industry for clinical application.

Gene Delivery

The introduction into a cell of a DNA sequence encoding for a therapeutic protein can be used to stimulate tissue repair or to have therapeutic effects in cardiovascular, cancer and metabolic disease. Chitosan can be used as a carrier to take DNA into the cell and achieve production of the protein. This approach avoids the immunological and oncogenic problems associated with viral introduction of foreign DNA but suffers from a lower level of production of the protein compared to the viral approach. We have leveraged our expertise in chitosan production, characterization and polyeletrolyte behavior to create chitosan/DNA nanoparticles that achieve very high levels of transgene expression. We are studying the mechanisms responsible for these high transfection levels using both physicochemical characterization of chitosan-DNA interactions and the cell biological events involved in the transfection process. Animal studies are also underway to investigate the use of these gene-bearing nanoparticles as therapeutic agents.

Biomechanics

The ultimate function of joint tissues such as cartilage, meniscus and bone is biomechanical. Articular cartilage, for example, must withstand and survive enormous pressures that can exceed 100 times atmospheric pressure and simultaneously provide joints surfaces that have almost no friction. The molecular structure and physical mechanisms that endow articular cartilage with these abilities are only partly elucidated. We develop new instruments that test the biomechanical properties of cartilage and its interesting electrical behavior where electric fields are produced inside cartilage when it is compressed. Mathematical models are also used to accurately predict the forces developed in cartilage under different loading conditions. These models account for the presence of a solid cross-linked macromolecular network and the pressurized fluid within it. The generation of electric fields in compressed cartilage is also measured using systems of microelectrodes. This latter technology has been translated to industry in the design of an instrument (http://www.biomomentum.com/Arthro-BST.html) that is being used to map the function of articular cartilage in patients undergoing arthroscopic surgery.
